摘  要:
为了筛选血清1型鸭甲型肝炎病毒(DHAV)衣壳蛋白的优势B细胞抗原表位及其各结构蛋白的最佳宿主表达系统,通过RT-PCR获得DHAV衣壳蛋白的编码基因(P1基因)序列,并分别运用DNAStar和EMBOSS软件分析其B细胞抗原表位区和密码子偏爱性特征,然后进行各结构蛋白抗原性的Western-bolt分析以及与大肠杆菌、酵母及人的密码子偏爱性进行比较。结果显示,DHAV的优势B细胞抗原表位主要存在于VP1和VP3蛋白,且每种蛋白各包含2个潜在的优势B细胞抗原表位。DHAV 3种结构蛋白基因的ENc值分别为54.631、49.502和45.924。VP3和VP1蛋白与酵母的密码子偏爱性差异最小,VP0蛋白与人的密码子偏爱性差异最小。结果表明,3种结构蛋白密码子出现的频率较为均一。VP1和VP3蛋白均是DHAV基因工程疫苗研制的候选免疫原,且适宜在酵母表达系统中表达,而VP0蛋白更适宜在人源细胞中表达。

摘  要:
To study the B cell epitopes and expression host system of capsid protein of duck hepatitis A virus(DHAV) serotype 1,the capsid protein-coding sequence(P1 gene) of DHAV was amplified by reverse transcription-polymerase chain reaction.The induced amino acid sequence of P1 of DHAV was analyzed with DNAStar software and EMBOSS database,and then identified with Western-blot and compared with codon bias of Escherichia coli,yeast and human.The results showed that the dominant B cell epitopes mainly presented in VP3 and VP1 protein of DHAV,and two dominant B cell epitopes were respectively involved in each protein.The ENc values of DHAV capsid proteins(VP0,VP3,VP1) were 54.631,49.502 and 45.924,respectively.Codon bias of VP3,VP1 and VP0 protein of DHAV were closest to that of yeast and human.The results indicated that the frequency of codon usage were basically consistent among the three structural proteins of DHAV.Both VP3 protein and VP1 protein were considered as the preferred immunogens and best expressed in yeast expression system.However,VP0 protein could be better expressed in human cell expression system.
