摘  要:
采用实时荧光定量PCR方法研究了生长速度不同的高邮鸭和金定鸭13、17、21、25、27胚龄和出雏后7日龄下丘脑中TRH基因mRNA的表达水平,并分析了其与胚重或体重的关系。结果表明:两鸭种在13胚龄时已均可检测到TRH基因mRNA的表达,从21胚龄到出雏后7日龄,TRH基因mRNA的表达量均一直增加,且出雏后7日龄的表达水平均显著高于胚胎期;两鸭种在胚胎期没有显著差异,而在出雏后7日龄时金定鸭极显著高于高邮鸭;相关性分析表明,两鸭种下丘脑中TRH基因mRNA的表达与胚重或体重均极显著正相关。研究结果提示,下丘脑中TRH基因mRNA的表达具有显著的年龄依赖性及组织差异性,出雏后还具有明显的品种特征,其对鸭早期生长发育可能有正调控作用。

摘  要:
By real-time PCR the thyrotrophin-releasing hormone(TRH) mRNA expressions in hypothalami were compared between Gaoyou ducks and Jinding ducks of different growth rates at embryonic stages of 13,17,21,25 and 27 days and age of 7 day,and their correlations to embryo weight and body weight were also analyzed.The results showed that the expressions of TRH mRNA in both duck breeds' hypothalami were detectable at 13-d embryonic stage(E13),and from E21 onwards the expression levels increased progressively and were significantly higher at the age of 7 day than at the embryonic stages;The expression levels were not significantly different between two breeds at the embryonic stages but at the age of 7 day the expression level of Jinding ducks was significantly higher than that of Gaoyou ducks.The correlation analysis showed that there was a very significantly positive correlation between both duck breeds' expression levels of TRH mRNA in hypothalami and their embryo weight or body weight.The above results indicated that the expression of TRH gene mRNA was affected obviously by age,breed and organization,and could have a positive regulatory effect on early-stage growth and development of ducks.
