对长豇豆品种ISSR反应体系中4种关键成分的优化

摘  要:
对长豇豆ISSR反应中模板、引物、dNTP和酶的浓度进行了初步筛选,通过正交设计实验确定了这4种关键成分的最终浓度,即引物为0.2μM、模板为0.015ng/μL、dNTP各200μM/L和Taq聚合酶为0.5U时,ISSR扩增效果最好。研究为利用ISSR分子标记技术鉴定长豇豆种质资源奠定了基础。

摘  要:
After the initial screening for the proper concentrations of template,primer,dNTP and enzyme,their ultimate concentrations were determined by an orthogonal design experiment.The best electrophoreses profile was produced when the concentrations of primer,template,dNTP and Taq polymerase were 0.2 M,0.015 ng/L,200 M/L and 0.5U,respectively.This study set up a foundation for the identification of asparagus bean germplasm with ISSR technique.
