康乐黄鸡TLR4基因的多态性分析及其与IL-2水平相关性的分析

摘  要:
为了研究康乐黄鸡Toll样受体4(TLR4)基因的多态性及其与白细胞介素-2(IL-2)水平的相关性,试验选取260只万载康乐黄鸡,翅下静脉采血,全血提取DNA并检测血清中IL-2浓度,对TLR4基因第三外显子区域进行扩增并测序,检测其单核苷酸多态性(SNP)位点,分析康乐黄鸡群体遗传变异情况以及SNP位点与血清中IL-2浓度的相关性。结果表明:所提取的基因组DNA完整,目的基因大小为517 bp,在线比对结果相似度达到了98%,可确定该目的基因为预计的基因片段,并且在TLR4基因的第三外显子处检测到SNP位点(C180T),CC和CT基因型频率显著高于TT基因型(P<0.05),该突变位点对康乐黄鸡血清中IL-2浓度有显著影响。说明可将C180T位点作为影响康乐黄鸡免疫指标IL-2的候选SNP位点进行进一步研究。

摘  要:
In the experiment, 260 blood samples of Wanzai Kangle yellow chickens under the subwinged veins were collected, DNA was extracted from the whole blood, and IL-2 concentration in the serum was detected. The exon 3 of TLR4 gene was amplified and sequenced to detect its single nucleotide polymorphism(SNP) site, and to understand the genetic variation of the healthy Kangle yellow chickens population and the correlation between the SNP-site and the concentration of IL-2 in serum.The results showed that the genomic DNA extracted by the test is complete, the size of the target gene was 517 bp, the similarity of online comparison results reached 98%. The target gene determined to be the expected gene fragment, and the SNP-site(C180 T) was detected at the exon 3 of of TLR4.The genotype frequency of CC and CT were both significantly higher than TT(P<0.05),and this mutation site had a significant effect on the serum IL-2 concentration of Kangle yellow chickens. It indicates that C180 T site can be used as a candidate SNP site that affects immune indexes of Kangle yellow chickens for further study.
