鸡蛔虫凉山州分离株线粒体ATP6基因的序列测定和种系发育分析

摘  要:
利用PCR技术,对分离自四川省凉山州17个鸡蛔虫分离株的线粒体ATP合成酶F0亚单位6基因(ATP6)的全序列进行扩增和分析。测序结果显示,所获得的鸡蛔虫ATP6全序列长度均为597bp,有25个变异位点,共检测到11个单倍型,种内变异为0.0~2.4%。种系发育树显示,所有鸡蛔虫分离株聚在同一分支,能与其他蛔虫相鉴别。结果表明,鸡蛔虫的ATP6基因序列种内变异小,种间差异大,故可作为分子标记用于鸡蛔虫的种间鉴定;鸡蛔虫凉山州分离株存在一定的遗传变异和遗传多样性。本研究结果为鸡蛔虫的分子分类、分子流行病学调查和种群遗传的进一步研究奠定了基础。

摘  要:
The complete sequence of mitochondrial ATP synthase F0 subunit gene(ATP6)of 17 Ascaridia galli samples collected from Liangshan,Sichuan province,was amplifed and sequenced.The results showed that the complete sequence of ATP6 gene of A.galli was 597 bp,with 25 variable sites.Eleven haplotypes were detected in these sequences,sequence variations within A.galli were 0.0-2.4%.The phylogenetic tree showed that all the A.galli isolates were clustered in the same clade,which can be identified with other Ascaridida.The results indicated that there was no significant variation in ATP6 sequences within A.galli,while there was a obvious difference interspecies,so ATP6 gene can be used as genetic marker to identify the roundworm of A.galli,and certain genetic variation and genetic diversity were presented among A.galli isoaltes from Liangshan.The results of this study provided the foundation for further research on molecular classification,epidemiological survey and population genetics of A.galli.
