摘  要:
【目的】阐明家蚕抗菌肽基因对病毒感染的中肠免疫应答模式,为揭示昆虫抗病毒机制提供参考。【方法】采用喂食家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,Bm NPV)给家蚕的方式进行诱导,以实时荧光定量PCR检测诱导后家蚕抗菌肽基因的表达情况,并比较Bm NPV诱导下8类已知家蚕抗菌肽主基因(cecropin D、moricin、gloverin2、defension B、attacin1、enbocin2、lysozyme和lebocin3)在肠道组织中表达水平的差异。【结果】在喂食Bm NPV处理3 h时,家蚕中肠组织中呈上调表达的抗菌肽基因仅有moricin基因;处理6 h时,cecropin D、gloverin2、moricin基因转录水平均呈明显的上调趋势,尤其以gloverin2基因的诱导活性相对最高,是对照组gloverin2基因的9.22倍;而在处理12和24 h时检测,发现8类已知家蚕抗菌肽基因表达量均小于1.0,呈下调趋势。【结论】Bm NPV侵染家蚕后gloverin2基因表达量明显上调,可能是gloverin2基因作为主要功能基因在病毒侵染早期过程中发挥了重要作用。

摘  要:
【Objective】The present experiment was conducted to investigate immune response patterns of antibacterial peptide genes to virus infection in midgut, in order to provide references for antiviral mechanism of insect. 【Method】The antibacterial peptide genes of Bombyx mori were induced by feeding on B. mori nucleopolyhedrovirus(Bm NPV). Then the expression of 8 kinds of major antimicrobial genes( cecropin D, moricin, gloverin2, defension B, attacin1, enbocin2,lysozyme and lebocin3) and their expression differences were analyzed by real-time fluorescent quantitative PCR technique. 【Result】The results showed that, after feeding on Bm NPV, the expression of only gene moricin was up-regulated at3 h, and the transcription levels of gene cecropin D, gloverin2, moricin were significantly increased at 6 h, especially gene gloverin2 with higher induced transcriptional activity which was 9.22 times than that of control group. However, the expression quantities of 8 kinds of antimicrobial genes in the midgut were less than 1.0 at 12 and 24 h, which showed down- regulated trend. 【 Conclusion 】 Significant up- regulated expression of glovin2 in B. mori infected with Bm NPV suggested that glovin2 gene can pay a role in the early stage of virus infection.
