北五味子藤茎总三萜对小鼠酒精性肝损伤的保护作用

摘 要:
目的:研究北五味子藤茎总三萜化合物(TFSCBR)对小鼠急性酒精性肝损伤的保护作用。方法:采用56°北京红星二锅头白酒建立急性酒精肝损伤模型。将60只昆明小鼠随机分为6组,每组10只,分别为正常对照组、模型对照组及TFSCBR高、中、低剂量组和齐墩果酸阳性对照组。每天灌胃1次,TFSCBR高、中、低剂量分别为50、30、10mg/(kg.d),齐墩果酸阳性对照组剂量为30mg/(kg.d),正常对照组、模型对照组灌胃等量生理盐水,实验4周后,通过脏器质量和体质量计算肝脏指数,测定小鼠血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)的含量,测定肝组织中过氧化脂质降解产物丙二醛(MDA)、还原型谷胱甘肽(GSH)和超氧化物歧化酶(SOD)含量。结果:中剂量的TFSCBR能显著降低酒精性肝损伤所引起的ALT、AST活性,抑制MDA含量的升高(P<0.01),有效地拮抗肝脏中SOD的活性与GSH含量的降低(P<0.01)。肝脏病理学改变减轻或逆转。结论:TFSCBR中剂量对酒精性肝损伤具有保护功能。

摘 要:
Objective: To explore the protective effect of triterpene compounds from Schisandra chinensis(Turcz) Baill rattan(TFSCBR) against acute alcohol liver injury in mice.Methods: Acute alcoholic liver injury model was induced by 56% Beijing Red Star Erguotou liquor.Totally 60 Kunming mice were randomly divided into 6 groups of 10 mice each: normal group,alcohol model group,oleanolic acid(positive control,30 mg/(kg.d)) group,and triterpenoid low,middle and high dose(10,30 mg/(kg.d) and 50 mg/(kg.d)) groups.The normal group and alcohol model group were administered with an equal volume of normal saline.After 4 weeks of administration,the levels of serum alanine transaminase(ALT),aspartate transaminase(AST) and liver malondialdehyde(MDA),glutathione(GSH) and super oxid edismutase(SOD) were assayed.Results: TFSCBR treatment at the dose of 30 mg/(kg.d) significantly reduced the elevation of AST,ALT and MDA(P < 0.01),and significantly inhibited the decrease of SOD activity and GSH level(P < 0.01) in mice suffering from liver injury.Furthermore,hepatic pathological changes were attenuated or even reversed.Conclusion: TFSCBR treatment at the dose of 30 mg/(kg.d) has a significant hepatoprotective effect against alcohol-induced liver injury in mice.
