利用mtDNA d-loop分析石迁野生鸳鸯在系统发育中的定位

摘  要:
采用PCR产物直接测序方法测定鸳鸯线粒体DNA(mtDNA)控制区的全序列,测定其与GenBank上已知雁形目28个物种的遗传距离,并构建系统发育树。结果表明,试验样品鸳鸯(采自贵州石阡县)mtDNA d-loop序列全长为1 035 bp,与欧洲鸳鸯(GenBank登录号AY112953)的遗传距离最近、为0.002,NJ分子系统树也显示两者同属一个分支,即鸳鸯为雁形目鸭科栖鸭族鸳鸯属,这与传统分类学方法一致。

摘  要:
Used direct sequencing of PCR product to determine Aix galericulata's mitochondrial DNA(mtDNA) control region sequences,measured genetic distance with the 28 species which belonged to Anseriformes as known in GeneBank and construct the phylogenetic tree.The results showed that the sample,which collected from Shiqian county of Guizhou province,Aix galericulata's mtDNA d-loop sequence is 1 035 bp.The nearest genetic distance between the sample mandarin duck and European mandarin duck(the GenBank accession number is AY112953) is 0.002.Constructed NJ molecular phylogenetic tree also showed they belonged to a branch,so the sample mandarin duck was belong to Aix Cairinini Anatidae Anseriformes,which was consistent with traditional taxonomy methods.
