17β-雌二醇对鸡胚原始生殖细胞体外培养的影响

摘  要：
为探讨17β-雌二醇(E2)对体外鸡胚原始生殖细胞(EPGC)发育的影响,采用EDTA-胰酶解离法获得第28期鸡胚性腺中的PGCs,依据差速贴壁原理分离纯化PGCs,然后置于添加不同浓度(0、5、10、20 ng/mL)17β-E2的培养液中进行培养。结果显示,5 ng/mL 17-βE2添加组与0 ng/mL 17-βE2对照组相比,PGCs的发育能力无显著差异,而10 ng/mL和20 ng/mL17β-E2两添加组与0 ng/mL及5 ng/mL组相比,PGCs的发育能力差异显著(P<0.05)。但是,10 ng/mL和20 ng/mL 17β-E2两添加组相比,PGCs的发育能力无显著差异。由此说明在体外环境中,一定浓度的17β-E2(10 ng/mL、20 ng/mL)可促进鸡原始生殖细胞的发育。

摘  要：
The effects of 17β-estradiol(E2) on development of chicken primordial germ cells(PGCs) in vitro were investigated.Primordial germ cells,which were isolated from genital ridge of stage 28 chicken embryos with the method of EDTA-tyrpsin collection,were purified by means of differential attachment technique and cultured in culture mediums of different 17β-E2 concentrations(0 ng/mL,5 ng/mL,10 ng/mL and 20 ng/mL).The results indicated that there was no significant difference in development of PGCs between the group with 0 ng/mL and 5 ng/mL 17β-E2.However,compared with the groups with 0 ng/mL and 5 ng/mL 17β-E2,the groups with 10 ng/mL and 20 ng/mL 17β-E2 had significant difference(P<0.05).But there was no significant difference in development of PGCs between the group with 10 ng/mL and 20 ng/mL 17β-E2.The findings indicate that 17β-estradiol of certain concentration(10 ng/mL,20 ng/mL) promotes PGCs development in vitro.
