兔小肠上皮细胞体外分离培养

摘  要:
选用新生未哺乳仔兔,应用胶原酶Ⅳ消化法对小肠上皮进行分离培养,得到兔小肠上皮细胞后,联合应用相差消化法和相差贴壁法对兔小肠上皮细胞进行纯化。通过形态学观察及细胞免疫组织化学方法对所得到的纯化兔小肠上皮细胞进行鉴定。结果表明:应用胶原酶Ⅳ消化法得到的兔小肠上皮细胞生长状况良好,纯化后得到95%以上纯度的兔小肠上皮细胞,纯化所得的兔小肠上皮细胞经形态学观察及细胞角蛋白8单克隆抗体鉴定为阳性。

摘  要:
The neonate rabbit was used in this experiment,and collagenase Ⅳ digestion was used to isolate and culture the IEC of rabbit.The rabbit IECs were purified by the way of phase difference digestion and adherence.And the IEC was identified with morphological observation and cyto-keratin detection,using mouse anti-cyto-keratin monoclonal antibodies.The result shows that the IEC by digesting with collagenase Ⅳ grew well.95% for the purification of the IEC was obtained.The result of identification by detecting the cyto-keratin was positive.
