海洋放线菌FA-F-5对水产病原菌的抑制作用

摘  要:
【目的】为了筛选可用于水生动物疾病防治的生防菌。【方法】以从福建琅琦岛海泥中分离得到的1株对多种水产病原菌具有较强抑制作用的菌株FA-F-5为研究对象,通过形态观察、生理生化测定、16SrDNA序列测定对其进行鉴定,并对其发酵条件和产生活性物质的理化性质进行了研究。【结果】初步判定菌株FA-F-5为弗氏链霉菌(Streptomyces fradiae);FA-F-5产生的抑菌活性物质的最佳发酵条件为:发酵培养基配方为麦芽糖15 g/L、黄豆粉25 g/L、CaCO31 g/L、K2HPO40.5 g/L,MgSO4.7H2O 0.5 g/L,海盐10 g/L,发酵培养基初始pH=8,培养温度25℃,种子液菌龄48 h,接种量100 mL/L,装液量100 mL/L,发酵时间7 d;该抑菌活性物质具有较好的热稳定性和pH稳定性;纸电泳试验证实该抑菌活性物质呈弱碱性。【结论】放线菌FA-F-5能够产生抑制多种水产病原菌的抑菌活性物质,有望开发成为新一代生物渔药的生防菌。

摘  要:
【Objective】The study is to screen the biocontrol agents against aquatic pathogens.【Method】FA-F-5 was isolated from marine sediment of Langqi Island in Fujian Province,which has strong activity against aquatic pathogens.The mycelium and spore morphology,physiological properities,16SrDNA sequence of FA-F-5 and optimal fermentation condition of this strain for the biosynthesis of bioactive metabolites were investigated.【Result】The results show that FA-F-5 was Streptomyces fradiae.The optional fermentation condition was proved that medium at pH=8,25 ℃,15 g/L maltose,25 g/L soybean powder,CaCO3 1 g/L,K2HPO40.5 g/L,MgSO4·7H2O 0.5 g/L,sea salt 10 g/L,inoculate with 48 hours' seed-age and 100 mL/L of inocula and culture for 7 days.It possesses fairly good heat and pH stabilities.Paper electrophoresis had proved that it was a substance of weak alkaloid.【Conclusion】FA-F-5 can produce active components to inhibit aquatic pathogens,which can be explored as one kind of fish biomedicines.
