五倍子提取物对1~42日龄黄羽肉鸡生长性能、肠道形态、免疫功能、抗氧化能力及肠道菌群的影响

摘  要:
本试验旨在研究饲粮添加五倍子提取物对1~42日龄黄羽肉鸡生长性能、肠道形态、免疫功能、抗氧化能力及肠道菌群的影响。选择600只1日龄初始体重为(39.75±0.02) g的健康新广901肉公雏,随机分为4个组,每组6个重复,每个重复25只。对照组饲喂基础饲粮,抗生素组在基础饲粮中添加2 mg/kg恩拉霉素,五倍子1组在基础饲粮中添加250 mg/kg五倍子提取物,五倍子2组在基础饲粮中添加500 mg/kg五倍子提取物。试验期42 d。结果表明:1)与对照组相比,五倍子2组42日龄体重、22~42日龄平均日增重和1~42日龄平均日增重均升高,但差异不显著(P>0.05)。2)与对照组相比,五倍子2组回肠绒毛高度和绒毛高度/隐窝深度值显著升高(P<0.05);且五倍子2组回肠绒毛高度/隐窝深度值显著高于抗生素组(P<0.05)。3)与对照组相比,五倍子1组和五倍子2组空肠分泌型免疫球蛋白A(sIgA)和γ-干扰素(IFN-γ)含量均显著升高(P<0.05),空肠白细胞介素-8(IL-8)和肿瘤坏死因子-α(TNF-α)含量均显著降低(P<0.05);且五倍子2组空肠sIgA含量显著高于抗生素组(P<0.05),空肠TNF-α含量显著低于抗生素组(P<0.05)。4)与对照组相比,五倍子1组和五倍子2组回肠丙二醛(MDA)含量显著降低(P<0.05),回肠总超氧化物歧化酶(T-SOD)活性显著升高(P<0.05)。5)与对照组相比,五倍子2组回肠闭合小环蛋白-1(ZO-1)和核因子E2相关因子2(Nrf2) mRNA相对表达量均显著升高(P<0.05);且五倍子2组回肠ZO-1和Nrf2 mRNA相对表达量显著高于抗生素组(P<0.05)。6)与对照组相比,五倍子1组和五倍子2组盲肠另枝菌属(Alistipes)和理研菌属(Rikenella)相对丰度显著降低(P<0.05),盲肠芽孢杆菌属(Bacillus)和毛螺菌属(Lachnospira)相对丰度均显著升高(P<0.05)。综上所述,饲粮添加500 mg/kg五倍子提取物可提高1~42日龄黄羽肉鸡生长性能、免疫功能和抗氧化能力,同时改善肠道形态结构、屏障功能和微生物菌群结构,可作为肉鸡抗生素潜在替代品。

摘  要:
The purpose of this study was to investigate the effects of gallnut extract on growth performance,intestinal morphology,immune function,antioxidant capacity and intestinal microbiota of yellow-feathered chickens during 1 to 42 days of age. A total of 600 healthy one-day-old female Xinguang 901 chickens with initial body weight of(39.75±0.02)g were randomly assigned to 4 groups with 6 replicates per group and 25 chickens per replicate. Chickens in the control group were fed a basal diet,chickens in the antibiotic group were fed the basal diet supplemented with 2 mg/kg enlanamycin,chickens in the gallnut 1 group were fed the basal diet supplemented with 250 mg/kg gallnut extract,and others in the gallnut 2 group were fed the basal diet supplemented with 500 mg/kg gallnut extract. The experiment lasted for 42 days. The results showed as follows:1)compared with the control group,the body weight at 42 days of age,average daily gain during 22 to42 days of age and average daily gain during 1 to 42 days of age of gallnut 2 group were increased,but had no significantly difference(P>0.05). 2)Compared with the control group,the villus height and villus height/crypt depth value in ileum of gallnut 2 group were significantly increased(P<0.05);and the villus height/crypt depth value in ileum of gallnut 2 group was significantly higher than that of antibiotic group(P<0.05).3)Compared with the control group,the contents of secretory immunoglobulin A(sIgA)andγ-interferon(IFN-γ)in jejunum of gallnut 1 group and gallnut 2 group were significantly increased(P<0.05),the contents of interleukin-8(IL-8)and tumor necrosis factor-α (TNF-α)in jejunum were significantly decreased(P<0.05);and the jejunal sIgA content of gallnut 2 group was significantly higher than that of antibiotic group(P<0.05),the jejunal TNF-αcontent of gallnut 2 group was significantly lower than that of antibiotic group(P<0.05). 4)Compared with the control group,the ileal malondialdehyde(MDA)content of gallnut 1 group and gallnut 2 group was significantly decreased(P<0.05),and the ileal total superoxide dismutase(T-SOD)activity was significantly increased(P<0.05). 5)Compared with the control group,the mRNA relative expression levels of zonula occludens protein 1(ZO-1)and nuclear factor-E2 related factor 2(Nrf2)in ileum of gallnut 2 group were significantly increased(P<0.05);and the mRNA relative expression levels of ZO-1 and Nrf2 in ileum of gallnut 2 group were significantly higher than those of antibiotic group(P<0.05). 6)Compared with the control group,the relative abundances of Alistipes and Rikenella in cecum of gallnut 1 group and gallnut 2 group were significantly decreased(P<0.05),and the relative abundances of Bacillus and Lachnospira in cecum were significantly increased(P<0.05). In summary,dietary supplemented with 500 mg/kg gallnut extract can improve the growth performance,immune function and antioxidant capacity of yellow-feathered chickens during 1 to 42 days of age,improve the intestinal morphology,barrier function and microflora structure,it can be used as a potential substitute for antibiotics in chickens.[Chinese Journal of Animal Nutrition,2022,34(3):1547-1558]
