新城疫病毒TS09-C耐热株灭活疫苗研究

摘  要:
选用β丙内酯对新城疫病毒TS09-C耐热株进行灭活处理,探索该毒株的最适灭活条件,并对灭活后病毒的耐热特性和保存期进行检测,同时制备油乳剂灭活疫苗接种2周龄健康SPF鸡,免疫后14d检测其新城疫抗体水平。结果显示,经0.25g/Lβ丙内酯37℃灭活2h,可彻底灭活TS09-C株,同时可保证其良好的HA血凝效价和耐热特性,灭活后的TS09-C株在42℃条件下保存60d,其HA效价未见显著下降,而灭活的La Sota非耐热株已经降为0。TS09-C株和La Sota株利用同样方法制备的油乳剂灭活疫苗免疫SPF鸡后产生的新城疫抗体水平分别为27.3和26.7。该研究为研发抗原稳定性强的新城疫灭活疫苗打下了良好的前期基础。

摘  要:
propiolactone was subjected to inactivate Newcastle disease virus thermostable TS09-C strain,the optimal inactivation conditions,thermostable characteristics and long time storage of the completely inactivated viruses were investigated.The two-week-old healthy SPF chickens were inoculated with the oil emulsion inactivated vaccine,and the anti Newcastle disease virus antibody titer was tested at 14 days post vaccination.The results showed that the TS09-C virus was completely inactivated by 0.25 g/Lβ-propiolactone under 37℃for 2 h.After inactivation,the virus remained the initial HA titer and thermostability.The HA titer of inactivated TS09-C virus reduced slightly after storage for 60 days at 42℃,while that of La Sota virus reduced to 0.Inactivated oil emulsion vaccine of thermostable TS09-C strain and thermolabile La Sota strain were prepared by the same method,and used to immunize SPF chickens.The average ND HI titers of TS09-C and La Sota vaccines were 27.3 and 26.7,respectively.The study provided a support for developing novel NDV inactivated vaccine with improved antigen stability.
