牛乳腺干细胞的分离和生物学特征

摘  要:
为获得牛乳腺干细胞并对其生物学特征进行研究,通过悬浮培养,从体外培养的乳腺上皮细胞分离得到乳腺干细胞球,并通过RT-PCR、Western-blot、免疫组化染色和流式细胞分析对其表面抗原标记特征和分化特征进行研究。结果表明,牛乳腺干细胞表达CD29和CD49f,可以分化为腺上皮细胞和肌上皮细胞。该分离方法增加了获得牛乳腺干细胞的途径,特异性标记物CD29和CD49f可用于其生物学特征研究。

摘  要:
To obtain bovine MaSCs and investigate the biological characteristics,mammary stem cell spheres were harvested by suspension culture from the mammary epithelial cells cultured in vitro,and their differentiation characteristics and special markers were identified by RT-PCR,western blotting,immunohistochemistry and flow cytometry analysis.The results showed that bovine MaSCs expressed CD29 and CD49f,and they could give rise to mammary epithelial cells and myoepithelial cells.It was suggested that employing this approach increased the access to isolate bovine MaSCs and the specific markers,CD29 and CD49f,could be used for the cell biological characteristics research.
