轮叶党参皂苷对二乙基亚硝胺致肝细胞DNA损伤的保护作用

摘  要:
目的:探讨轮叶党参皂苷(Codonopsis lanceolata saponin,CLS)对二乙基亚硝胺(DEN)致肝细胞DNA损伤的保护作用,为开发肝癌化学预防剂提供参考依据.方法:将50只小鼠随机分为5组:阴性对照组(每日灌胃蒸馏水10mL/(kg·d)),DEN组(隔日腹腔注射DEN 20mg/kg),CLS高、中、低剂量组(每日分别灌胃CLS 200、100、50mg/(kg·d),同时隔日腹腔注射DEN 20mg/kg),连续给药6周.采用单细胞凝胶电泳技术分别对肝细胞DNA损伤进行分析,用比色法测定肝组织中丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活力.结果:CLS对DEN引起小鼠体质量增长抑制作用没有明显的改善作用.CLS 3个剂量组的彗星尾长、彗星长、尾矩、Olive尾矩及尾DNA百分率均显著低于单纯DEN组(P<0.05),各剂量组DNA损伤程度均低于DEN组(P<0.05).CLS高剂量组肝组织SOD活性明显高于DEN组,CLS高、中剂量组肝组织GSH-Px活性明显高于DEN组(P<0.01),CLS 3个剂量组肝组织匀浆中MDA含量均显著低于DEN组(P<0.01),且呈剂量依赖性减少.结论:轮叶党参皂苷对DEN致小鼠肝细胞DNA损伤有一定的保护作用.

摘  要:
Objective: To explore the preventive effect of total saponin(CLS) from Codonopsis lanceolata on DNA damage in liver cells induced by diethylnitrosamine(DEN),and provide a reference for chemical prevention of liver cancer.Methods: Fifty mice were randomly divided into five groups including negative control group(intragastric administration of distilled water),DEN group(DEN 20 mg/kg by intraperitoneal injection every other day),and high,middle and low dose groups(CLS 200,100 mg/(kg.d) and 50 mg/(kg.d) by intragastric administration every day,DEN 20 mg/kg by intraperitoneal injection every other day).The treatments were lasted for 6 weeks.DNA damage of liver cells were analyzed by a single cell gel electrophoresis technique,the content of malondialdehyde(MDA),the activities of glutathione peroxidase(GSH-Px) and superoxidedismutase(SOD) in liver tissues were determined by colorimentry.Results: CLS had no significant improvement for mice on the inhibition of body weight.Comet tail length,comet length,tail moment,Olive tail moment and tail DNA(%) of three CLS dose groups were significantly lower than those of DEN group(P < 0.05),DNA damage of each CLS dose group was significantly lower than that of DEN group(P < 0.05).SOD activity of CLS high dose group was significantly higher than that of DEN group(P < 0.01),GSH-Px activities of CLS high and middle groups were significantly higher than that of DEN group(P < 0.01),the contents of MDA in three CLS dose groups were significantly lower than that of DEN group in a dose-dependent manner(P < 0.01).Conclusion: Codonopsis lanceolata saponin has obvious protective effects against DNA damage in mouse liver cells induced by DEN.
