摘 要:
采用生物信息学方法系统分析了甜菜夜蛾、菜粉蝶、烟夜蛾和黑腹果蝇泛素延伸蛋白(UBE)基因序列的遗传多样性、分子进化及其编码氨基酸序列的结构功能域和基序特点。结果显示,4种昆虫UBE基因序列的同源性较高(81.8%~99.7%),且遗传距离较近(0.003~0.199),均编码ubiquitin/L40 UBE。共检出81个多态位点,生成4个单倍型,单倍型多样性(Hd=1.000)、平均核苷酸差异数(K=45.883)、核苷酸多样性(Pi=0.112 97)、同义替换核苷酸多样性均值(Pi(s)=0.415 01)、非同义替换核苷酸多样性均值(Pi(a)=0.024 89)、密码子有效值(ENC=38.070~53.458<61)、偏爱指标(CBI=0.437~0.729>0)和χ2检验计算值(χ2=0.454~0.960)表明这4种昆虫UBE基因的遗传多样性较丰富且呈现较强的密码子偏爱性。4条氨基酸序列均含泛素同系物结构域和Ribosomal/L 40结构域各1个,未见信号肽和跨膜结构域;共含14条基序,其中Se-UBE和Dm-UBE基因编码的氨基酸序列各具1个酪蛋白激酶Ⅱ磷酸化位点和2个蛋白激酶C磷酸化位点,Pr-UBE和Ha-UBE基因编码的序列则各含2个酪蛋白激酶Ⅱ磷酸化位点和2个蛋白激酶C磷酸化位点。研究结果为进一步开展UBE基因分子进化机制、表达特性及昆虫病毒侵染关系等方面的研究提供基础信息。

摘 要:
Based on the published sequences of ubiquitin extension protein(UBE) genes of Spodoptera exigu,Pieris rapa,Helicoverpa assulta,Drosophila melanogaste in NCBI database,the genetic diversity and molecular evolution of the nucleotide sequences,as well as structural domains and motif characters of putative amino acid sequences were analyzed using the bioinformatics method.Results showed high identity(81.8%-99.7%) and short genetic distances(0.003-0.199)between the four UBE gene sequences,and all encoded ubiquitin/L40 UBE.During the genetic diversity analysis,81 polymorphic sites in total were detected from 4 UBE gene sequences and 4 haplotypes were sorted.Abundant genetic diversity and strong codon bias were found based on the haplotype diversity(Hd=1.000),the average number of nucleotide difference(K=45.883),the nucleotide diversity(Pi=0.11297),the nucleotide diversity of synonmyous sites(Pi(s)=0.41501),the nucleotide diversity of nonsynonmyous sites(Pi(a)= 0.02489),the effective number of codons(ENC=38.070-53.458<61),the codon bias index(CBI=0.437-0.729>0) and the scaled Chi-square(χ2=0.454-0.960).Each amino acid sequence deduced from its corresponding UBE gene contained one ubiquitin homologue and one Ribosomal/L40 domain,respectively,but signal peptide and transmembrane domain were not found.Fourteen motifs were detected among 4 amino acid sequences.Deduced amoino acid sequences from the Se-UBE gene and Dm-UBE gene contained 1 casein kinase Ⅱ phosphorylation site and 2 protein kinase C phosphorylation sites.Deduced amino acid sequences from the Pr-UBE gene and Ha-UBE gene both had 2 casein kinase Ⅱ phosphorylation sites and 2 protein kinase C phosphorylation sites.The result provided some basic information for further studying the evolution and expression characteristics of UBE genes as well as mechanism of insect viral infection.
