转绿色荧光蛋白基因家蚕品系的纯化与综合经济性状改良

摘  要:
为了使转绿色荧光蛋白基因家蚕品系能在生产上推广应用,利用传统育种方法对转基因家蚕品系进行系统分离纯化和综合经济性状的杂交改良。对转基因家蚕材料西绿的绿色荧光茧性状进行分离纯化后,按茧色分成深绿茧品系、中绿茧品系、浅绿茧品系,其中深绿茧品系的绿茧率达100%,但在纯化过程中发现外源基因转入对家蚕的茧形与茧层率、丝质等经济性状均有不良影响。以纯化后的转基因家蚕西绿的深绿茧品系为父本,分别与普通白茧家蚕实用品种932、7532进行杂交,经多代选育获得具有一定生产性能的转基因家蚕绿茧品系,该转育品系的平均虫蛹生命率在90%以上,茧色纯化快,茧形正,缫丝成绩优于西绿。

摘  要:
In order to use transgenic silkworm(Bombyx mori) line with green fluorescent protein gene for production,we used traditional breeding methods to separate and purify the transgenic silkworm line and employed crossbreeding to improve its comprehensive economic traits.After green cocoon trait of a transgenic silkworm line named Xilü was separated and purified,three strains named dark green cocoon,mid-green cocoon and light green cocoon were discriminated based on their cocoon colors and the green cocoon rate of dark green cocoon strain reached 100%.However,during the purification process,it was found that introduction of exogenous gene had adverse effect to cocoon shape,cocoon shell rate,silk quality and other economic traits.Therefore,the purified dark green strain of transgenic silkworm line Xilü was used as the male parent to hybridize with two silkworm practical varieties 932 and 7532 which spin normal white cocoons.After multi-generational selection and breeding,a transgenic green cocoon silkworm strain with good productivity was obtained.Average larva-pupa survival rate of this hybrid strain exceeded 90%.Besides,this strain had a high purification rate on cocoon color,a good cocoon shape and better reeling data than Xilü.
