摘 要:
为明确甘肃某羊场发生的疑似绵羊肺炎支原体肺炎的病原,利用支原体培养基从一份患肺炎的绵羊肺脏病料中分离到一个菌落。通过菌落形态观察、生化试验和特异性PCR方法进行了鉴定。结果分离物在固体培养基上生长的菌落呈圆形无中心脐,能水解葡萄糖但不能水解精氨酸,不消化酪蛋白,PCR能扩增出绵羊肺炎支原体特异的大小为361bp的目的片段,测序比对表明该基因序列与绵羊肺炎支原体标准株Y98的同源性为98%。鉴定结果表明引起该养殖场发病的病原为绵羊肺炎支原体。

摘 要:
To identify the pathogen from sheep showed pneumonia symptoms occurred in a sheep farm in Gansu province,one isolate was isolated from one pneumonic lung tissue sample of sheep using mycoplasma medium.The mycoplasma isolate was characterized by the morphologic observation,biochemistry test and specific PCR test,colonies of isolate grown on solid medium were typical un-oil omelette-like,isolate can hydrolyze glucose but cannot hydrolyze arginine,and digest casein,a specific target fragment of 361 bp can be amplified by PCR,sequence analysis of the 16 S rRNA gene revealed that the identity is 98% with standard strain of Y98.The results indicated that the isolate belongs to Mycoplasma ovipneumoniae,which will provide help for the farm.
