摘  要:
鸡白细胞介素12(ChIL-12)是由IL-12p40和IL-12p35两个亚基组成的一个异源二聚体,具有促进T淋巴细胞和自然杀伤细胞(NK cell)成熟的作用,能够作为黏膜疫苗佐剂。本研究中采用公认的食品级乳酸乳球菌表达系统构建表达目的融合蛋白Ch IL-12p40-(G_4S)_3ChIL-12p35(rChIL-12),并经western blot检测。结果显示:目的融合蛋白呈可溶性状态表达。体外鸡淋巴细胞刺激试验显示,经rChIL-12处理的鸡脾淋巴细胞能够产生干扰素γ(IFN-γ),表明该融合蛋白具有诱导IFN-γ产生的生物学活性。本研究结果为进一步利用rChIL-12作为粘膜疫苗佐剂的研发奠定了基础。

摘  要:
Chicken interleukin-12(ChIL-12) is a heterodimeric cytokine consisted with IL-12p40 and IL-12p35 subunits.ChIL-12 has the ability to promote the mature of T cells and natural killer cells(NK cell), which is used as a mucosal vaccine adjuvant. In this study, the IL-12p40 and IL-12p35 subunit genes were fused with a glycine-serine linker encoding sequence by PCR, and cloned into the plasmid of pNZ8149 for expression in the food-grade NZ3900 strain of Lactococcus lactis. Western blot detection showed that the recombinant Ch L-12 fusion protein(rChIL-12) was expressed in a soluble form in the cytoplasm.Moreover, the r ChIL-12 was able to induce chicken splenic mononuclear cells to produce interferon-γ verified by the chicken lymphocyte stimulation test in vitro. These results demonstrated that the rChIL-12 has the biological activity, which laid the foundation for further study of r ChIL-12 as a mucosal vaccine adjuvant.
