小麦和荞麦过敏原的分离与免疫学特性初步鉴定

摘  要:
为了提取、分离小麦(Triticum aestivum)和荞麦(Fagopyrum esculentum)的主要过敏原并对其免疫学特性进行鉴定,以磷酸盐缓冲液(PBS)制备小麦和荞麦总蛋白粗提液,通过SDS-PAGE分析总蛋白组成,利用麦类食品过敏患者血清中的IgE能特异性结合过敏原的特性,通过免疫印迹(Western-Blotting)鉴定其中的过敏原成分,然后用离子交换层析(DE-52)对总蛋白进行初步分离,并用Western-Blotting鉴定分离后组份的免疫学特性。SDS-PAGE分析显示,小麦蛋白条带共有18条,其中主要条带有12条(55、42、39、36、29、27、23、21、18、15、12、9 kD);荞麦蛋白条带共有17条,其中主要条带有8条(55、44、36、32、22、16、13、10kD)。Western-Blotting鉴定表明,小麦阳性条带有9条(36、32、29、26、23、18、15、12、9 kD),其中23 kD的过敏原特异性最强;荞麦阳性条带有5条(55、36、32、292、2 kD),其中22 kD的过敏原特异性最强。DE-52阴离子交换层析分离后经Western-Blotting分析显示,分离后过敏原的相对含量均有显著提高,且依然具有较强的免疫学活性,如经纯化后荞麦22 kD的过敏原相对含量由10.73%提高到21.07%。初步鉴定出小麦的主要过敏原为23 kD蛋白,而荞麦的主要过敏原为22 kD蛋白。

摘  要:
To extract,isolate and identify the main protein allergens from wheat(Triticum aestivum)and buckwheat(Fagopyrum esculentum),the total protein from wheat and buckwheat were extract by PBS,the allergens were identified with 10 allergic patients' serum by Western-Blotting after SDS-PAGE.To partially purify the allergens,Ion exchange chromatography technique was employed.Finally immune activeness of the partially purify allergens was identified by means of Western-Blotting.In the crude extract from wheat,there are eighteen protein bands visible on the SDS-PAGE gel,and the most abundant 12 bands are 55 kD,42 kD,39 kD,36 kD,29 kD,27 kD,23 kD,21 kD,18 kD,15 kD,12 kD,9 kD proteins,respectively.Similarly,in the crude extract from Buckwheat,there are seventeen protein bands visible on the SDS-PAGE gel,and the most abundant 8 bands are 55 kD,44 kD,36 kD,32 kD,22 kD,16 kD,13 kD,10 kD proteins,respectively.The allergens from wheat with molecular mass of 36 kD,32 kD,29 kD,26 kD,23 kD,18 kD,15 kD,12 kD,9 kD were identified by Western-Blotting,and the primary allergen is 23 kD protein.The allergens from Buckwheat with molecular mass are 55 kD,36 kD,32 kD,29 kD,22 kD were identified by Western-Blotting.The primary allergen is 22 kD protein.The content of buckwheat allergens was raised from 10.73% to 21.07% after DE-52 anion exchange chromatography.Allergens from Triticum aestivum and Fagopyrum esculentum were isolated and identified.The result will provide a basis for the diagnosis and treatment of allergy to the allergens from Triticum aestivum and Fagopyrum esculentum.
