2种车轮虫DNA抽提方法比较

摘  要:
[目的]比较研究了2种抽提车轮虫DNA的方法,苯酚-氯仿法与试剂盒抽提法。[方法]采用苯酚-氯仿法与试剂盒抽提法对采自重庆地区淡水鱼外寄生的纤细小车轮虫进行了DNA抽提比较研究。通过琼脂糖凝胶电泳技术和核酸蛋白检测技术对所抽提的DNA及PCR扩增产物进行了分析。[结果]苯酚-氯仿法抽提的DNA中杂质含量较少,但需要的车轮虫个体数量多;而试剂盒则可有效地抽提极微量的车轮虫DNA,对于无法进行单克隆培养的生物或无法得到足量标本的种类,则具有更好的应用前景。[结论]在试验过程中可以根据研究的实际情况因地制宜地选择使用不同的方法,以达到高效、经济的目的。

摘  要:
[Objective] The research aimed to study the 2 methods on DNA extraction of Trichodinids subtilisa Lom.,the phenol-chloroform method and the SIGMA RED Extract-N-Amp Tissue PCR Kit method.[Method] Two methods,the phenol-chloroform method and the SIGMA RED Extract-N-Amp Tissue PCR Kit method,were used to extract DNA for the research of small subunit(SSU) rRNA gene of Trichodinids subtilisa Lom.isolated from freshwater fishes in Chongqing.Extracted DNA and PCR products were analyzed and used to compare the effect of the two methods by the agarose gel electrophoresis and detection of spectrophotometer(U-0080D).[Result] The results revealed that DNA of Trichodinids subtilisa Lom.extracted by phenol-chloroform method was of less impurity,but needed more Trichodinids subtilisa Lom.samples.However,the SIGMA RED Extract-N-Amp Tissue PCR Kit could also be used to extract DNA of Trichodinids subtilisa Lom.fast and efficiently with less quantity,which had given a bright future for these species that were uneasy to acquire or culture clonally.[Conclusion] Different method can be chose according to different practical situation during experiment,to realize efficient and economic purpose.
