IFN-γ对妊娠早期大鼠下丘脑中IL-18表达的影响

摘  要:
为探讨γ干扰素(IFN-γ)对妊娠早期大鼠下丘脑中白细胞介素-18(IL-18)表达及其对妊娠结局的影响,将妊娠9d的SD大鼠随机分为对照组、试验1组和试验2组,采用生殖道肌肉注射的方法,对照组大鼠生殖道肌肉注射生理盐水,试验组大鼠注射IFN-γ,其中1组剂量为2.5×104 U/只,2组剂量为7.5×104 U/只。各试验组均在注射48h后灌注取材、免疫组化SP法染色,结果表明,IL-18在下丘脑视前室周核、视上核、视前大细胞核、视前内侧核、视前外侧核均具有较多数量的阳性神经元表达;阳性细胞呈圆形、卵圆形、三角形不等,阳性物质大多位于细胞质和细胞核。生殖道肌肉注射2.5×104 U及7.5×104 U的IFN-γ后能下调IL-18在下丘脑表达的同时会造成妊娠终止。提示IFN-γ对妊娠早期大鼠下丘脑中IL-18的表达具有调节作用,且外源注射7.5×104 U IFN-γ后会造成妊娠早期大鼠流产。

摘  要:
In order to investigate the effect of γ-interferon(IFN-γ) on the expression of interleukin-18(IL-18) in the hypothalamus and pregnant result of pregnant rat,earlier pregnant rat(D9),were divided into control group,experiment group 1 and experiment group 2.The rats in control group were injected with physiological brine,and the rats in other two groups were injected with 2.5×104 IU IFN-γ and 7.5×104 IU IFN-γ respectively in muscle of vagina.The perfusions of each group were collected 48h after injection.Hypothalamus of rats in each group was collected and stained by immunohistochemical SP method.The result showed as follows:there were many IL-18 positive expression in nucleus pre-opticus medianus,nuclens supra-opticus,nuckeus pre-opticus mango cellularis,nucleus pre-optius medialis and nucleus pre-opticus lateralis in the hypothalamus.The positive neurons were in round,oval round and triangle.And the positive neurons were mainly distributed in plasma and nucleus.The injection of 2.5×104 IU and 7.5×104 IU IFN-γ by vaginal muscle could reduce the expression of IL-18 and stop pregnant.The results suggested that the IFN-γ has regulatory effect on the expression of IL-18 in the hypothalamus,IL-18 on pregnancy and the regulation of IFN-γ has a very close relationship.
