阿霉素诱导兔心衰模型的建立与评价

摘  要:
探讨静脉注射阿霉素(ADR)建立兔心衰动物模型的可行性。将日本大耳白兔32只,随机分成模型组1、2和对照组;模型组1注射ADR 1mg/kg,每周2次,共8周;模型组2注射ADR 2mg/kg,每周1次,共8周;对照组注射相同体积9g/L NaCl注射液。于停药3周后进行心脏彩超检查,并进行心肌组织病理切片观察其组织学改变。与对照组相比,模型组动物左室舒张末期容积明显增大,左室射血分数明显降低(P<0.05),病理检查符合扩张性心肌病样改变;模型组间无统计学差别。常规剂量多次静脉注射ADR可显著损伤心肌,造成心脏舒缩功能下降,是建立动物心衰模型的一种简便可行的方法。低浓度多次注射更有利于制模期间的观察。

摘  要:
To investigate the feasibility of the rabbit heart failure animal model by injecting intravenous adriamycin(ADR).32Japanese white rabbits were randomly divided into Model group 1,2and Control group,Model group 1:injection of ADR 1mg/kg,2times in a week,for 8weeks;model group 2:injection of ADR 2mg/kg,once in a week,for 8weeks;control group:injected with the same volume of 9g/L NaCl injection.After three weeks of withdrawal,rabbits in the three groups were measured by color doppler ultrasound;and the myocardial tissue sections were examined for pathological lesions.Compared with the control group,the left ventricular end-diastolic volume of model groups increased significantly,LVEF was significantly lower(P<0.05),conforming pathological changes of cardiomyopathy,and the two model group were not statistically different.By conventional dose multiple injection,ADR could injure significantly myocardial organization and reduce cardiac ventricular and systolic functions.This is a simple and practical method to establish animal heart failure model.Low concentrations of multiple injections were more conducive in observation of model establishment.
