西部白眉长臂猿支气管败血波氏杆菌的分离鉴定及其耐药基因检测

摘  要:
为探究某动物园西部白眉长臂猿死亡的可能原因,无菌采集病死白眉长臂猿肺脏组织进行细菌分离鉴定。通过分离纯化、培养特性观察、革兰染色镜检、生化试验以及16S rDNA克隆测定对分离菌株进行鉴定;进一步研究分离株的致病性、耐药性,并进行超广谱β-内酰胺酶耐药基因检测及测序分析。结果表明,分离得到1株革兰阴性小杆菌,命名为J181121,该分离菌的培养及生化特性符合支气管败血波氏杆菌,其16S rDNA基因序列与Genbank中支气管败血波氏杆菌(登录号:CP018761.1)的同源性为99%,对小鼠致病力强。药敏试验结果显示,分离株对β-内酰胺类抗生素耐药,PCR检测到超广谱β-内酰胺酶耐药基因bla-SHV。

摘  要:
In order to explore the possible death cause of the Hylobates hoolock in a zoo, the bacteria was isolated and identified from the lungs. Isolation and purification, culture characteristics observation, gram staining microscopy, biochemical test and 16 S rDNA cloning were conducted to identify the isolated strains. The pathogenicity and drug resistance of the isolates were further studied, and the ultrabroad spectrum beta-lactamase resistance genes were tested and sequenced. The result showed that a strain of gram-negative bacillus was successfully isolated, which was named J181121. The isolate was tested for pathogenicity and drug resistance, and further extended-spectrum β-lactamase resistance genes were detected and sequenced. The culture and biochemical characteristics of J181121 were consistent with Bordetella bronchiseptica, and the 16 S rDNA gene sequence of this isolate showed 99% homology with B. bronchiseptica(accession number: CP018761.1) in genbank. And it was highly pathogenic to mice. The drug sensitivity test showed that the isolate was resistant to β-lactam antibiotics, and the extended-spectrum β-lactamase gene bla-SHV could be amplified by PCR.
