嘉兴黑猪ESR2和FSHβ及PRLR基因多态性与繁殖性状相关性分析

摘 要:
本研究旨在检测嘉兴黑猪ESR2、FSHβ和PRLR基因的多态性及其对繁殖性能的影响。通过聚合酶链式反应-单链构象多态性分析(PCR-SSCP)检测嘉兴黑猪273个个体8个单核苷酸多态性(SNP)位点的多态性,并与母猪繁殖性能(总产仔数、产活仔数、死胎数、两胎间隔和初生均重)开展关联分析。结果表明:ESR2基因外显子1的A221G与母猪的总产仔数、产活仔数、死胎数和初生均重显著相关(P<0.001),PRLR基因的A13901T影响总产仔数、产活仔数、死胎数和初生均重(P<0.001);上述2个位点合并基因型分析表明基因型为AA-TT和AGTT的个体繁殖性状显著高于其他部分基因型个体(P<0.001);其余位点并未发现与繁殖性状显著关联(P>0.05)。本研究与常用的ESR2、FSHβ和PRLR基因酶切位点分析不同,筛选并确定了8个嘉兴黑猪新的SNP位点,其中2个位点和繁殖性状紧密相关,这为提高嘉兴黑猪种群的遗传育种水平和生产繁殖能力提供了理论支撑。

摘 要:
To investigate the effects of ESR2, FSHβ, and PRLR genes on reproductive traits of Jiaxing Black pig, we used PCR-SSCP to examine the genotypes of eight SNPs in 273 individuals of this breed, and analyzed correlation between genotypes and five reproduction-related parameters(total number of piglet born(TNB), number of piglet born alive(NBA), number of stillborn pigs(NSP), farrawing interal(FI), and litter weight at birth(LWB)). The results suggested A221 G of ESR2 gene was related with TNB, NBA, NSP, and LWB(P <0.001); A13901 T of PRLR gene affected TNB,NBA, and LWB(P<0.001); and the individuals with AA-TT(A221G and A13901 T loci) and AG-TT combined genotypes have significantly higher reproductive traits than others(P<0.001). Compared with the common enzyme digestion loci in ESR2, FSHβ, and PRLR genes, we determined new SNP locus, two of which were closely related to reproductive traits.These results provide a theoretical support for improving the breeding selection of this population.
