小麦醇溶蛋白酸性聚丙烯酰胺凝胶电泳方法的优化及应用

摘  要:
为了形成一套适合实际情况的小麦醇溶蛋白酸性聚丙烯酰胺凝胶电泳(A-PAGE)方法和谱带分析技术;依据国际种子检验协会1986年公布的标准小麦醇溶蛋白A-PAGE方法,结合国内实验室的实际情况,摸索出了适合中国试剂和仪器的小麦醇溶蛋白A-PAGE的改良方法。该方法克服了标准方法中的胶韧性差、剥胶困难、分辨率不高、谱带识别繁琐、实验效率较低的限制。该方法适合中国小麦品种纯度的快速鉴定、血缘关系分析、遗传育种种质材料的选择等。

摘  要:
This paper is to formulate a set of acid polyacrylamide gel electrophoresis of wheat gliadin and the analytical technique of the bands suiting our actual conditions.Based on the standard acid polyacrylamide gel electrophoresis of wheat gliadin was published by ISTA in 1986,combined with the actual circumstance of national laboratories,and explore the improved methods of the acid polyacrylamide gel electrophoresis of wheat gliadin of the reagents and equipment for China.This method overcame the many restrictions of the standard method,such as the poor gel toughness,the gel desquamated uncomfortably,low resolution,troublesome identification of the bands,less efficiency of the experiment.This method suited the rapid identification and the consanguinity analysis of the purity of the Chinese wheat breed and the choice of the germplasms in genetic breeding and so on.
