长白山中华蜜蜂基因组CBS-700(bp)DNA同源片段克隆及其Southern杂交的研究

摘  要:
本文通过分析12个长白山中蜂(长白山地方采样点中华蜜蜂)基因组多态性分布规律,以寻找其基因组特异性分子标记为主要目的;以RAPD-PCR技术选择性扩增基因组RAPD位点分子,以克隆有意义的分子标记,DNA酶Ⅰ将克隆的分子标记酶学合成为探针并进行Southern杂交等为主要方法;结果发现1005号蜂等10个基因组分子标记多态性图谱同时显示700(bp)DNA片段,而1083号蜂等2个基因组分子标记多态性图谱仅为弱显示或未显示700(bp)DNA片段;这一结果提示:700(bp)DNA片段可能是长白山中蜂基因组有意义的分子标记,即保守的同源序列,可以充当长白山中蜂基因组的特异性分子标记。

摘  要:
[OBJECTIVE]The article was about sorting specific molecular biomarker by analyzing polymorphism map of genome of local Changbaishan Apis cerana ceranas;[METHOD]primarily using the method of RAPD-PCR for selectively amplifying molecular markers of polymorphism from genomes of 12 local groups to clone representative molecular biomarker , and then synthesizing probe of DNA fragment by DNAⅠfollowed with Southern hybridization [RESULTS]was that the 10 local groups (represented especially as No1005) showed in common bands 700(bp) in the polymorphism map of genome. As opposed to the 10 groups, 2 local groups(represented especially as No1083) showed bands 700(bp) in faintness or did not show .[CONCLUSION]indicated that bands 700(bp)of fragment DNA ,homogeneous DNA, was preliminarily attributed to the genetic and could maybe be acted as specific molecular biomarker .
