粗野鹿角珊瑚产纤维素酶细菌的筛选、鉴定和酶活测定

摘 要:
为获得高效降解纤维素活性菌株,采用富集平板稀释法、刚果红染色法筛选粗野鹿角珊瑚纤维素分解菌,分离得到2株产纤维素酶细菌HZXJ007和HZXJ010,经16S rDNA序列分析,HZXJ007与产气肠杆菌只同源性达99%以上;HZXJ010与紫金牛叶杆菌同源性高达99%以上。采用DNS法测定菌株降解纤维素酶活,HZXJ007和HZXJ010的滤纸酶活分别为197.67、210.55 U/mL,内切酶(C x)活分别为79.42、86.83 U/mL,外切酶(C1)活分别为87.47、104.87 U/mL,β-葡萄糖苷酶活分别为47.52、46.88 U/mL。试验结果表明,两菌株酶系齐全活性强,在以木质纤维素为原料生产纤维素乙醇方面有潜在应用价值。

摘 要:
To obtain high-efficient strain for degradation cellulose, two cellulase, producing bacteria HZXJ007 and HZXJ010, were isolated from Acopora Humilis Dana after using concentration and plates serial dilution methods and Congo red staining method; The homogeneity of HZXJ007 with Enterobacter aerogenes appeared to be 99% and HZXJ010 shares99% homology with Phyllobacterium myrsinacearum by 16s rDNA sequence analysis; DNS method was used for determinating enzyme activity of strains. Their filter paper enzyme activities were 197.67 U/mL and 210.55 U/mL, Cx activies were 79.42 U/mL and 86.83 U/mL, c1activies were 87.47 U/mL and 104.87 U/mL, the β-glucosidase activity were47.52 U/mL and 46.88 U/mL, respectively. The results showed that the two strains could produce high and complete degradation cellulose enzymes, so they had potential application value in the process of cellulosic ethanol production with raw lignocellulose material.
