鸡microRNA基因“seed”区4个SNPs的群体遗传分布

摘  要:
采用CRS-RFLP技术研究了rs14076349、rs16681032、rs14934924和rs16681031共4个SNPs位点在蛋肉兼用型、蛋用型和药用观赏型3种经济类型的6个鸡群的遗传多样性。结果表明:rs16681032在所有鸡群中未检测到变异,rs16681031、rs14934924和rs14076349位点具有较高的次等位基因频率,分别为0.4553、0.1173和0.3296。兼用型鸡与药用观赏型鸡在rs16681031位点的基因频率分布差异显著(P<0.05);兼用型鸡与蛋用型鸡在rs14934924位点的基因频率分布差异极显著(P<0.01);rs14076349位点在所有经济类群间的基因频率分布差异极显著(P<0.01)。根据基因频率信息进行聚类分析,大致可将6个鸡群分为3类,聚类结果反映出不同鸡种microRNAs调控通路的差异。研究结果表明,鸡种系特异性microRNA基因可能受到正选择的影响而存在加速进化现象,其"seed"区SNPs位点在不同鸡种的经济表型的形成中可能发挥着重要作用。

摘  要:
To investigate the evolution pattern of chicken-specific microRNAs and the genetic distribution difference of SNPs located in seed regions among three economic groups of chicken.Genetic diversity of rs14076349,rs16681032,rs16681031 and rs14934924 among six chicken populations which belong to egg meat dual purpose,egg type and medicinal and ornamental chickens were determined using created restriction site PCR-RFLP method.The results showed that no variation was detected in rs16681031 sites in six chicken populations.The rs16681031,rs14934924 and rs14076349 also showed a greater minor allele frequency.The results also showed that an extreme significant differences in the frequency of the rs14076349 site among three economic groups of chicken(P<0.01).Egg meat dual purpose chickens had significant differences in the frequency of the rs16681031(P<0.05)and medicinal and ornamental chickens.Egg type chickens had an extreme significant differences in the frequency of the rs14934924(P<0.01)and egg meat dual purpose chickens.The dendrograms based on allele frequency divided the six chicken populations into three clusters, which reflected the difference of regulatory pathway of microRNAs in six chicken populations.The results suggested that positive selection and evolutionary acceleration has occurred in the seed regions of chicken-specfic microRNAs during chicken evolution.And these SNPs located in the seed regions of chicken-specfic microRNAs may play an important role in the formation of economic phenotype of different chicken breeds.
