新扬州鸡IGF-Ⅰ基因多态性的PCR-SSCP检测及其与生长发育性状的相关性

摘  要:
以新扬州鸡胰岛素样生长因子-Ⅰ(IGF-Ⅰ)基因作为控制其生长、发育性状的候选基因,通过对IGF-Ⅰ基因主要部分序列进行PCR-SSCP检测,发现了2个多态位点.测序结果表明,IGF-Ⅰ基因存在2个突变位点:插入(G)和(G→C).分析多态位点与0-12周龄鸡生长发育性状的关联性,结果表明:插入(G)突变基因型中,2周龄鸡体重BB型与AB型差异显著(P<0.05),但AA型与AB型和BB型差异均不显著,而(G→C)突变各基因型间没有显著差异;插入(G)突变位点对0-12周龄鸡的龙骨长、胫骨长均没有显著影响,而(G→C)突变位点仅在鸡6周龄时表现出与龙骨长、胫骨长间的差异显著性.

摘  要:
Due to the importance of insulin-like growth factor-Ⅰ(IGF-Ⅰ) in regulating many physiological and metabolic processes in animal body,we applied PCR-SSCP to detect the mutation of IGF-Ⅰ gene in New Yangzhou chickens and to analyze DNA sequence.We gained two mutation loci of IGF-Ⅰ gene: insert (G),(G→C).The association between DNA mutation loci and chicken traits was analyzed with general line model.The results were as follows.In the insert(G) mutation genotypes,there was significant difference between body weights of 2 weeks aged BB and AB genotype(P<0.05),while there was no significant difference between AA and other genotypes.In the(G→C) mutation genotypes,there was no significant difference in body weight.Insert(G) mutation loci had no significant effect on fossil bone length and shank length of 0-12 weeks aged chickens,only(G→C) mutation loci had a significant effect on fossil bone length and shank length of 6 weeks aged chickens.
