改进的CTAB法提取蔓茎堇菜基因组DNA

摘  要:
研究了蔓茎堇菜(VioladiffusaGing.)基因组DNA的分离提取方法。在细胞核裂解之前,除去细胞质中的多糖等杂质,然后采用CTAB法分别从野生植株叶片、组培植株叶片、愈伤组织及悬浮培养细胞4种新鲜材料中成功地分离提取了蔓茎堇菜基因组DNA。所提取的DNA,链长度均大于48kb,OD260/OD280比值为1.8~1.9,产率为219.8~300.5μg/g,可直接用于限制性内切酶酶切,并适用于RAPD分析。

摘  要:
Polysaccharides and other secondary metabolites in the cytoplasm were removed before thenucleus was decomposed by CTAB, and the genomic DNA of Viola diffusa Ging. was isolated andpurified from leaves of wild plant, leaves of test-tube plant, calli and cells in suspension culture. Thelength of the isolated DNA fragment from different tissues was over 48 kb and the proportion ofOD260/OD280 of DNA was 1.8  ̄ 1.9. The yield of the DNA with this method was 219.8  ̄ 300.5 μg/g andthese DNA samples were all suitable for digesting with restriction enzyme and RAPD analysis.
