扁豆DNA提取方法的研究

摘  要:
以白花扁豆、眉豆和紫边扁豆3个品种为试材,对影响扁豆DNA提取质量的主要因子进行研究,以期建立适宜扁豆DNA提取的优化反应体系。结果表明,扁豆不同品种DNA提取中,紫边扁豆DNA质量较好;不同CTAB浓度的对比中,2%CTAB提取缓冲液提取DNA的效果较好;叶片和果实DNA提取中,扁豆叶片提取的DNA质量优于扁豆果实;CTAB和SDS两种提取方法对比,CTAB法提取出的DNA较纯,污染少。DNA提取优化后的结果为2%CTAB法提取的紫边扁豆叶片的DNA质量好,纯度高。

摘  要:
White flower dolichos lablab,vigna cylindrical and purple boundary dolichos lablab were used as experimental materials.The main factors affecting extraction quality of DNA from dolichos lablab were studied in order to establish an optimal system of DNA extraction.The results indicated that the DNA examination quality was quite good from purple boundary dolichos lablab;In the different CTAB density,2% CTAB extraction buffer was better;The leaf and fruit DNA extraction,the leaf was better than the fruit;the contrast of the CTAB and the SDS methods,DNA which the CTAB method to extracted were purer and less pollution.The optimi DNA extraction result was the leaf of purple boundary dolichos lablab and 2% CTAB extraction buffer.
