生物菌剂对樱桃的促生效应及根际细菌群落的影响

摘  要:
为探讨根际促生菌B20对樱桃的促生作用及对根际土壤细菌群落的影响,以发酵好的菇渣(ZT)为吸附载体,吸附B20菌株(JZ)制成生物菌剂(JF),对比研究了JZ和JF对樱桃的生长、根际土壤理化性质及土壤酶活性的影响。利用高通量测序技术分析研究了其对樱桃根际细菌群落的影响。结果表明:JF处理可以促进马哈利樱桃的生长,并能显著提高其干重和N、P和K养分含量。JF处理的地上部和根系干重分别比JZ处理增加178.10%和58.68%;根系P和K含量分别增加73.73%和40.21%。与对照相比,JF处理的土壤脲酶和蔗糖酶活性显著提高。JF处理显著增加了根际土壤中养分含量,速效磷和有效钾含量分别比对照增加73.07%和53.48%,但碱解氮含量差异不显著。高通量测序结果显示,共获得了941416条有效序列、20676个OTUs。多样性分析结果表明,JZ和JF的施用可提高樱桃根际土壤细菌群落结构的多样性及丰富度。对细菌群落组成分析发现,供试样品中的优势菌门均为变形菌门、酸杆菌门、厚壁菌门、放线菌门及拟杆菌门。经JF处理后,厚壁菌门所占比例明显增加,与对照相比,Bacillus,Lactobacillus含量由0.18%和0.05%增加到0.73%和0.62%。而Alistipes,Bacteroides及Lachnospiraceae_NK4A136_group是JF中特有的属种,在其他样品中没有检测到。由此可见,施用促生菌B20菌液和混配的生物菌剂均可影响马哈利樱桃根际土壤的细菌群落,促进樱桃的生长和养分吸收,但是混配的生物菌剂效果明显优于单独的促生菌液。

摘  要:
In order to determine the effects of plant growth promoting rhizobacteria on mahaleb growth and rhizosphere bacterial community, the biological agent(JF) was prepared by compounding dominant bacteria liquid B20(JZ) and decomposed mushroom residue(ZT). The effects of JF, JZ and ZT on growth, soil physical and chemical properties and soil enzyme activities were studied in pot experiment. The effects on the rhizosphere bacterial community structure were also investigated by the Illumina Hiseq high-throughput sequencing. The results indicated that JF not only promoted the absorption of nitrogen, phosphorus and potassium, but also increased the dry weight of mahaleb compared with blank medium(CK). Compared with JZ, the dry weight of aerial part and root in treatment JF increased by 178.10% and 58.68%, P and K nutrient uptake of root in treatment JF increased by 73.73% and 40.21%, respectively. Compared with the CK, JF significantly increased the activities of soil urease and sucrose. JF significantly enhanced the nutrient content of phosphorus and potassium by 73.07% and 53.48%, respectively,compared to than CK. However, it had no effects on available nitrogen content. We obtained a total of 941416 effective sequences and 20676 OTUs. The soils bacterial diversity and abundance increased after JZ and JF treatment. Bacterial community composition analysis showed that the dominant phyla in four soil samples were Proteobacteria, Acidobacteria,Firmicutes, Actinobacteria and Bacteroidetes. Compared with the CK, the ratio of Firmicutes increased in JF treatment soil, and the ratio of Bacillus, Lactobacillus increased from 0.18% and 0.05% to 0.73% and 0.62%, respectively. The ratio of Alistipes,Bacteroides and achnospiraceae_NK4 A136_group in JF were 1.56%, 2.52% and 0.86%, respectively. These species were not identified in the other samples. So, both the B20 bacterial liquid and biological agent could affected the rhizosphere bacterial community, and promote mahaleb growth and nutrient absorption. However, the effect of biological agent was better than the B20 bacterial liquid.
