摘  要:
根据GenBank中发表的I型鸭病毒性肝炎病毒(DHV-I)的3D基因设计一对引物,用实验室分离培养的毒株提取病毒RNA进行RT-PCR扩增,得到预期大小的目的片段。试验结果表明,所建立的RT-PCR方法最低模板检出量为50pg,且该方法只能特异地检测出鸭病毒性肝炎病毒,不能检出鸭瘟病毒(DPV)、禽流感病毒(AIV)和番鸭呼肠孤病毒(MDRV)等。该方法也能成功检测出人工感染发病死亡雏鸭的肝脏和脾脏。结果表明,该方法可用于DHV-Ⅰ的分离鉴定、临床病料的检测和分子流行病学调查。

摘  要:
According to the DHV-I gene which was published in genebank ,primers were designed with help of Primer Premier 5.0 design software,and a RT-PCR assay for the detection of DHV-Ⅰwas developed.A specific 277 bp fragment was amplified from DHV-I which is keeping in laboratory. The sensitivity and specificity of the RT-PCR method were high ,which could detect 50 Pg RNA of the DHVI,but couldn't detect DPV,AIV,MDRV. And it also can detect RNA from liver and spleen of dead ducks which were experimentally infected with DHV-Ⅰvirus. So the method can be used as identification of DHV-I,detection of the clinical samples and molecular epidemiological survey.
