摘  要:
白枕鹤(Grus vipio)和丹顶鹤(Grus japonensis)是中国重点保护动物,为单态性鸟,很难通过外观和形态来进行性别鉴定,给人工繁殖造成很大困难。研究已发现CHD基因和EE0.6序列片段大小在非平胸鸟类W、Z染色体上均存在差异。基于此,本研究以无锡市动物园饲养的白枕鹤和丹顶鹤为研究对象,经PCR实验,对白枕鹤和丹顶鹤CHD基因和EE0.6序列的相关片段进行了特异性扩增。结果显示,这2种方法 PCR扩增结果一致,并根据这一结果进行白枕鹤、丹顶鹤配对,均成功孵化出后代,表明该PCR鉴定结果可靠,研究结果为提高鹤类人为配对指数和繁殖率奠定了基础。

摘  要:
White-naped crane( Grus vipio) and red crowned crane( Grus japonensis) are national protected species in China. They are sexually monomorphic species and it is difficult to identify gender based on their external morphology. This causes difficulties in artificial propagation. Studies have found that there are sequence fragment size difference in the CHD and EE0. 6 genes on the W and Z chromosome of non-ratite birds. In this study, the fragments of CHD and EE0. 6 genes of white-naped and red-crowned cranes at Wuxi City Zoo were amplified by PCR. The results of two methods were consistent and were used to pair cranes of both species. The pairings resulted in successful production of offspring. The results of PCR gender determination were reliable. This study laid a foundation for increasing the artificial pairing index and reproduction rate of captive cranes.
