摘 要:
嗜热菌β-糖苷酶(lac S)由于具有催化糖苷键水解进而分解乳糖的作用,因而被应用于转基因牛的研究中。本研究利用构建的普通载体plox-EGF-lac S和含有转座元件的乳腺特异性表达嗜热菌β-糖苷酶载体ZGL-LACS-EN转染牛胎儿成纤维细胞,并将挑选出的阳性细胞用作供体细胞,构建转基因克隆胚,并比较两种转基因克隆胚的发育潜能,并进行胚胎移植。结果表明,普通载体转染的转基因胚胎卵裂率(83.2%)稍高于转座子介导的转基因胚胎(75.7%),而囊胚率(15.3%/14.9%)无明显差别,但两者均明显低于非转基因的胚胎(卵裂率88.2%,囊胚率36.7%)。这为进一步培育低乳糖奶牛新品种的研究奠定了基础。

摘 要:
Sulfolobus Solfataricus β-Glycosidases has a function on catalytic hydrolysis glycosidic bond and hydrolysis of lactose.This study has been successfully constructed common plox-EGF-lac S vector and ZGL-LACS-EN vector of containing transposable element and specific expression of Sulfolobus Solfataricus β-Glycosidases,in order to compare the difference gene embryos transfected by two vectors.Result showed that,the embryo cleavage rate of plox-EGF-lac S is slightly higher than ZGL-LACS-EN(83.2% /75.7%),and the blastosphere rate was no obvious differences between them(15.3% /14.9%),but non-modified embryos was obvious higher than both of them(cleavage rate 88.2%,blastosphere rate 36.7%).The above study do an exploratory research at cultivating of low lactose dairy cow new species.
