四个意大利蜜蜂品系RAPD遗传标记的分析

摘  要:
前期研究中筛选到可在平湖浆蜂 (Ep)、美意 (Em)、澳意 (Eo)和苏意 (Es)中扩增出多态性条带的RAPD -PCR随机引物W18(5’ -CGGACGGCGG - 3’)和W2 3(5’ -GTACCGCCCG - 3’)。本文以群体遗传学方法对这 4个意蜂品系的RAPD多态性扩增条带进行分析。分析结果表明 ,在引物W18扩增出的多态性条带中 ,2 35 2bp和 36 4bp条带仅在美意中检测到 ,其基因频率分别为 0 .0 5 5 9和 0 .0 4 2 0 ;6 32bp条带在平湖浆蜂中出现的基因频率 (0 .90 86 )远高于美意 (0 .0 36 3)、澳意 (0 .0 190 )和苏意 (0 .0 0 0 0 ) ;2 0 92bp条带在美意与澳意中出现的基因频率分别为 0 .0 2 2 3和 0 .0 6 6 7,而在苏意和平湖浆蜂未检测到该条带。在W2 3引物扩增条带中 ,仅在澳意中检测到 6 5 1bp条带 ,其基因频率为 0 .0 348。RAPD多态性扩增条带基因频率的聚类分析结果表明 ,平湖浆蜂与苏意的亲缘关系最近 ,其遗传相关系数为 0 .982 0 ;平湖浆蜂、苏意与美意的遗传系数均为 0 .970 0 ,而与澳意的遗传相关系数均为 0 .96 90 ,表明这两个地方蜂种与美意的亲缘关系略高于澳意。美意与澳意的亲缘关系相对较远 ,其遗传相关系数为 0 .96 90。本研究结果在鉴定意蜂地理亚种下的各品系中具有实际应用价值。

摘  要:
In the previous study, we reported2 primers, W18(5’-CGGACGGCGG-3’)and W23(5’-GTACCGCCCG-3’), could generated polymorphic bands in RAPD-PCR analysis of genomic DNA of 4 lines of A.m.ligusica (Pinghu royal jelly bee, E p ; American-Italian bee, E m ; Australia-Italian bee, E o and Jiangsu local Italian bee, E s ) In this article, those RAPD-PCR results were analyzed by population genetics methods. It was show that In the W18 primer amplified polymorphic bands, 2 fragments with the length of 2353 bp and 364 bp were detected only in E m ,their gene frequencies were 0.0559 and 0.0420, respectively. The 632 bp amplified band occurred at a much higher frequency in high royal jelly performance variety E p (0.91) than the other 3 lines (E m ,0.04; E o , 0.02 ; E s , 0.00) . Another polymorphic band with the length of 2092bp was detected only in E m and E o , and their gene frequencies were 0.0223 and 0.0667. In the W23 amplified bands, the 651bp band was amplified only in E o , with the gene frequency of 0.0348. Clustering analysis was further performed according to the gene frequencies of the RAPD polymorphic bands. It was showed that the genetic relative coefficient between E s and E p ,that was R( Es, Ep ) = 0.9820. was higher than the other 5 genetic relative coefficients(R( Es, Em ) = R( Ep, Em =0.9700,R( Es, Eo ) = R( Ep, Eo =0.9690,R( Em, Eo ) = 0.9690). This result indicated thatgenetic relationship of was closest in among the 4 Italian honeybee lines. The results of this research provided practical tool for identifying bee lines of geography subspecies A.m.ligusica.
