艾纳香遗传多样性的SRAP和AFLP对比分析

摘  要:
【目的】分析艾纳香的遗传多样性,为制定其保护策略提供参考依据。【方法】使用分子标记中常用的SRAP和AFLP分子标记对35份艾纳香资源的遗传多样性进行对比分析与评估。【结果】8对AFLP引物组合和27对SRAP引物组合分别扩增获得1360条AFLP多样性条带(多样性比率为99.48%)和265条SRAP多样性条带(多样性比率为97.78%);SRAP分子标记的有效信息位点指数(PIC)、有效多样性指数(EMR)和标记指数(MI)分别为0.4381、8.1000和4.3141,AFLP分子标记的PIC、EMR和MI分别为0.1773、198.0000和301.1348;基于AFLP多样性条带和SRAP多样性条带的遗传相似度对比分析结果表明,35份艾纳香样品的遗传相似度为0.4450~0.9179,两种分子标记的相关系数r=0.47;基于SRAP分子标记遗传相似系数,35份艾纳香样品可分为五大类群;而基于AELP分子标记遗传相似系数,35份艾纳香样品可分为四大类群。【结论】AFLP和SRAP分子标记均可用于艾纳香的遗传多样性分析,但AFLP分子标记分析的多样性位点多、分子标记特征指数高,更适合用于艾纳香的遗传多样性研究。

摘  要:
【Objective】The genetic diversity of Blumea balsamifera D C. was estimated, in order to provide reference basis for its protection strategy. 【Method】Based on AFLP and SRAP molecular markers, the genetic diversity of 35 B. balsamifera accessions was estimated. 【Result】A total of 1360 AFLP polymorphism bands(polymorphic ratio of 99.48%)and 265 SRAP polymorphism bands(polymorphic ratio of 97.78%) were amplified using 8 AFLP primer combinations and 27 SRAP primer combinations. Polymorphism information content(PIC), effective multiplex ratio(EMR) and marker index(MI) of SRAP molecular marker were 0.4381, 8.1000 and 4.3141, respectively. PIC, EMR and MI of AFLP molecular marker were 0.1773, 198.0000 and 301.1348, respectively. Based on AFLP and SRAP polymorphic bands, the genetic similarity of 35 accessions ranged from 0.4450 to 0.9179, and the correlation coefficient(r) was 0.47. The UPGMA clustering results showed that, based on genetic similarity coefficient of SRAP molecular marker, 35 accessions could be classified into 5 clusters, but based on genetic similarity coefficient of AFLP molecular marker, 35 accessions could be classified into 4 clusters. 【Conclusion】Both SRAP and AFLP molecular markers can used to estimate genetic diversity of B. balsamifera, but AFLP molecular marker is more suitable to estimate genetic diversity of B. balsamifera because it has more polymorphic sites and higher marker characteristic index.
