一株传染性造血器官坏死病毒的分离鉴定和系统进化分析

摘  要:
本研究从河北省某虹鳟养殖场取患病虹鳟组织样本,进行病毒分离、回归感染、RT-PCR检测及系统进化分析。分离鉴定出1株传染性造血器官坏死病毒(Infectious hematopoietic necrosis virus,IHNV),命名为IHNV-BJSY株。取病鱼组织匀浆滤菌液接种到鲤上皮瘤细胞(Epithelioma papulosum cyprini,EPC),出现典型的细胞病变效应(Cytopathic effect,CPE);健康虹鳟鱼苗腹腔注射病变细胞液回归感染出现急性死亡,10 d累计死亡率达72%,症状与自然发病鱼相同;RT-PCR检测显示自然发病鱼、病变细胞液和攻毒后死亡虹鳟样品均为IHNV阳性。糖蛋白基因核苷酸序列的系统进化分析结果显示,该病毒株与我国XJ-13株、GS2014株的同源性为99%,属于JRt基因型。

摘  要:
In this study, samples of diseased rainbow trout tissue were collected from a a rainbow trout farm in Hebei province for virus isolation, regression infection, RT-PCR detection and phylogenetic analysis. An infectious hematopoietic necrosis virus isolate named IHNV-BJSY was isolated. The tissue homogenate filtrate of diseased rainbow trout was inoculated into the carp epithelioma papulosum cyprinid(EPC) cells, and the typical cytopathic effect was formed. The healthy rain trout fry was injected intraperitoneally with infected cells liquid. The artificially infected rain trout fry were died acutely with a cumulative mortality of 75% 10 days after infection, and showing the similar clinical symptoms with the fish infected naturally. RT-PCR identification revealed that the simple from the fish naturally infected, the infected cells and artificially infected rain trout fry were all IHNV-positive. The phylogenetic analysis of the glycoprotein gene sequence showed that the IHNV-BJSY isolated had 99% identity of G gene sequence with Chinese isolate IHNV-XJ-13 or GS2014, belonging to the genotype JRt.
