摘  要:
自药用植物苦参中分离得到具有良好抗病效果的内生真菌BS002菌株,对该菌株发酵培养基的条件进行了探究。在单因素试验基础上,应用Plackett-Burman设计,得出主要影响因素有结晶乙酸钠、葡萄糖、马铃薯的含量;采用响应面分析法借助Box-Behnken试验设计,对内生真菌BS002菌株发酵条件进行优化,确定最佳条件为:装液量100 mL/250mL锥形瓶,马铃薯246.47 g/L,葡萄糖27.81 g/L,结晶乙酸钠1.95g/L,接种量10~7 cfu/mL,温度25℃,酵母膏1.0g/L,转速150r/min,时间4d,菌丝干重可达10.398 g/L。

摘  要:
The endophytic fungus BS002 with a good disease-resistant effect was isolated from medicinal Sophora flavescens,and its fermentation medium conditions were studied.Based on the monofactorial experiments and the Plackett-Burman design test it was concluded that the crystalline sodium acetate,potato and glucose contents were main influencing factors! The fermentation conditions of endophytic fungus BS002 were optimized by response surface methodology and the BoxBehnken design test,and the gained optimal conditions were as follows:100 mL solution in a 250-mL conical flask,246.47 g/L potato,27.81 g/L glucose,1.95 g/L crystalline sodium acetate,10~7 cfu/mL pitching rate,and 1.0 g/L yeast extract paste at 25 ℃ and 150 r/min speed for 4 d,in which the mycelium dry weight could reach 10.398 g/L.
