观赏凤梨DNA提取方法研究

摘  要:
观赏凤梨(Bromelia ceae)叶片呈革质化且多纤维,含大量次生代谢产物,常常导致提取的DNA产量低、质量差,影响后续的酶切和PCR扩增。针对观赏凤梨多个属的植物,通过多种不同因素包括提取液、提取部位、研磨方法、沉淀时间等的对比,对观赏凤梨的基因组DNA提取方法进行研究,成功地掌握了高效的凤梨叶片DNA提取方法,为凤梨后期分子标记等基因组研究打下一定的基础。

摘  要:
The leaves of ornamental Bromelia plants were coriaceous and rich in fibre and secondary metabolism compounds,which lead the DNA extraction unefficient and low quality,and influence the following enzyme digestion and PCR amplification.We chose several genuses of Bromelia plant,use different part of the matierals,compared with different buffer solutions,trituration methods and deposition time,and summarized a efficient method of DNA extraction for Bromelia plants.
