一株野生有柄树舌的鉴定、菌丝培养特性及驯化栽培

摘  要:
以一株野生大型真菌为研究对象,通过ITS(internal transcribed spacer)测序和同源序列比对鉴定该野生真菌,同时使用固体平板培养和人工栽培试验来探讨该野生真菌的培养特性等。结果表明:所分离野生真菌为有柄树舌(Ganoderma gibbosum),菌丝生长所需要的最佳碳源为葡萄糖或果糖,氮源为酵母浸膏,最适碳氮比为(10∶1)~(20∶1),pH 5.5,培养温度28℃。木屑、玉米芯和棉籽壳为主料的3种栽培基质中都观察到有柄树舌子实体的发生。

摘  要:
A wild macrofungi was taken as research object.The wild macrofungi was identified by internal transcribed spacer(ITS)sequence and homologous sequence alignments,simultaneously mycelial cultivation characteristics and artificial cultivation were investigated by solid plate culture method and artificial logs.The results showed that the isolating wild strain was Ganoderma gibbosum by phylogenetic analysis based on ITS region sequence.Optimum carbon was the glucose or fructose,and the optimum nitrogen source was extracts of yeast,and the optimum carbon nitrogen ratio was about from 10∶1to 20∶1.The optimum growth temperature and optimum pH value were around 28℃ and5.5respectively.The fruit body development was observed from three kinds of cultivating substrate whose main materials were sawdust,corncob and cottonseed hull separately.
