戊糖片球菌HS2细胞制备γ-氨基丁酸的研究

摘  要:
采用戊糖片球菌(Pediococcus pentosaceus)HS2细胞转化L-谷氨酸一钠制备γ-氨基丁酸(γ-aminobutyric acid,GABA)。通过对转化反应温度、pH值、金属离子、底物浓度、菌体浓度和菌体培养时间等因素对HS2细胞转化法生产GABA的影响进行了考查,确定转化反应体系最优的组成为:采用培养12h的菌体、湿菌体50mg/mL、Ca2+50mmol/L、L-谷氨酸一钠10g/L和L-谷氨酸50g/L。当该体系在pH值4.5、35℃、120r/min搅拌下反应48h,转化液中GABA的浓度达到(39.89±2.12)g/L,摩尔转化率为(97.00±5.16)%。

摘  要:
The bioconversion of L-glutamic acid to γ-aminobutyric acid (GABA) by the strain of Pediococcus pentosaceus HS2 was studied and the effects of temperature, pH value, metal ions, substrate concentration, cell concentration and cultivation time were investigated in this paper. The optimal composition of bioconversion system were 50 g/L concentration cell with 12h cultivation in 50mmol/L Ca2+, 10g/L monosodium L-glutamate and 50 mg/mL L-glutamic acid medium. The content of GABA reached (39.89±2.12) g/L after 48h fermentation at pH 4.5, 35℃ and 120 r/min, and GABA mole yield of bioconversion reached (97.00±5.16)%.
