高效液相色谱法测定葡萄中噻苯隆残留量

摘  要:
[目的]建立葡萄中噻苯隆残留量的液相色谱分析方法。[方法]样品经乙腈提取,盐析后上层有机相水浴60℃氮吹至近干,用甲醇定容后过有机滤膜供仪器检测。分析采用Kromasil C18色谱柱,甲醇、水作为流动相梯度洗脱,流速1.0 mL/min,检测波长290 nm,外标法定量。[结果]噻苯隆在0.05~1.0 mg/L质量浓度范围呈良好的线性,线性相关系数大于0.9999,方法检出限为0.02 mg/kg,定量限为0.05 mg/kg。添加质量分数0.05、0.5、1.0 mg/kg时,回收率为102.6%~111.2%,批内相对标准偏差为1.58%~4.01%(n=5),批间相对标准偏差为5.64%~6.23%(n=5)。[结论]该方法简单、快速、准确度高,适用于葡萄中噻苯隆的残留量测定。

摘  要:
[Aims] A high performance liquid chromatography was established for the determination of thidiazuron in grapes. [Methods] The samples were extracted with acetonitrile. The upper organic phase after salting-out was evaporated to dryness in a water bath at 60 ℃ under nitrogen and redissolved with 5 mL methanol. The thidiazuron in solution after filtrating with organic film was analyzed by HPLC-DAD, and a mobile phase of methanol-water. Detection wavelength was 290 nm and flow rate of the mobile phase was 1.0 mL/min. [Results] There was a good range of linearity within the range of thidiazuron concentration from 0.05 to1.0 mg/L. The linear correlation coefficient was > 0.999 9. The limit of detection(LOD) of the method was 0.02 mg/kg. The limit of quantification(LOQ) was 0.05 mg/kg. The recovery was102.6-111.2% in grapes. The within- and between-run relative standard deviations(RSDs) were 1.58-4.01% and 5.64-6.23%, respectively. [Conclusions] The simplicity, high-speed and good accuracy of the method made it be well suitable for determination of thidiazuron residue in grape.
