猪FOXO3基因的多态性和生长性状关联分析

摘  要:
为了探讨猪FOXO3基因SNPs位点及其与猪生长性状的关联性,采用基因克隆并测序的方法检测FOXO3基因的多态位点,并对其与生长性状进行关联分析。结果显示:该基因共存在3个SNP位点,为:G82316A、T60086C和A64011G,分别命名为:P1、P2和P3。在群体中均表现出3种基因型,3个位点对生长性状的影响表现为:P1位点AA基因型个体校正背膘厚显著高于AG基因型个体(P<0.05)以及GG基因型个体(P<0.05);P2位点CC基因型个体背膘厚显著高于TT基因型个体(P<0.05)以及TC基因型个体(P<0.05);P3位点AG基因型个体100 kg日龄显著低于AA基因型个体(P<0.05)以及GG基因型个体(P<0.05)。结论:P1、P2变异位点对大白猪的背膘厚及生长速度具有负选择作用,P3位点对大白猪的背膘厚和生长速度具有正选择作用。此3个多态位点可作为猪分子遗传育种的选择性标记。

摘  要:
To investigate SNPs in FOXO3 gene and the correlation between the SNPs and growth traits of pigs. Gene cloning and sequencing were adopted to investigate the SNPs, and its association with growth traits were analyzed. The result showed that a total of three SNPs were detected in FOXO3 gene, such as G82316 A, T60086 C and A64011 G,they were respectively named P1, P2, P3. The three sites were all expressed as three genotypes. The influence of the three sites on growth traits showed that the corrected back fat thickness and the age of reaching 100 kg of pigs with AA genotype were significantly higher than the ones with AG genotype(P<0.05) and the ones with GG genotype(P<0.05) in P1; the back fat thickness and the age of reaching 100 kg of pigs with CC genotype were significantly higher than the ones with TT genotype(P<0.05) and the ones with TC genotype(P<0.05) in P2;the back fat thickness and the age of reaching 100 kg of pig with AG genotype was significantly lower than the ones with AA genotype(P<0.05) and GG genotype(P<0.05) in P3. P1 and P2 sites display negative selection effects on back fat thickness and growth rate; P3 site displays positive selection affact the back fat thickness and growth rate.Therefore the sites can be selected as assistant-selected genetic markers for pig breeding.
