日照麻鸡ATPATP5A1W基因的克隆和生物信息学分析

摘  要:
ATP5A1W(ATP synthase subunit alpha)基因定位于鸟类W染色体上,其编码产物参与氧化磷酸化,在跨膜质子动力势的推动下合成ATP。本试验以日照麻鸡为试验材料,采用RT-PCR技术克隆ATP5A1W基因,结合生物信息学方法分析其生物学特征。结果表明,克隆得到日照麻鸡ATP5A1W基因c DNA长度为1 695 bp,开放阅读框1 662 bp,编码553个氨基酸。该核苷酸序列与Gen Bank(登录号:XM_429118)上预测的核苷酸序列一致性为100%。进化树分析表明,日照麻鸡ATP5A1W氨基酸序列与绿头鸭和鹅的进化关系较近。鸡ATP5A1W蛋白亚细胞定位于线粒体(78.3%)、细胞质(8.7%)、细胞核(8.7%)和细胞外(4.3%)。预测鸡ATP5A1W蛋白二级结构由42.86%的α-螺旋,19.89%的延伸链,10.49%的β-转角,26.76%的无规则卷曲组成。此研究结果将为开展日照麻鸡ATP5A1W基因的结构和功能提供参考。

摘  要:
The α subunit of mitochondrial ATP synthase(ATP5 A1 W) is located within the W chromosome of birds,which participates in oxidative phosphorylation and synthesizes ATP under transmembrane proton motive force. At present study, ATP5 A1 W gene was cloned by RT-PCR and analyzed biological characteristics of ATP5 A1 W gene by bioinformatics methods. The results showed that ATP5 A1 W partial c DNA of the Rizhao partridge chicken was 1 695 bp in length containing 1 662 bp open reading frame, encoded a protein with 553 amino acids. The identity of nucleotide sequences of Rizhao partridge chicken ATP5 A1 W shared 100% with predicted sequences of Gen Bank(ACCESSION:XM_429118). Phylogeny tree analysis indicated that amino acid sequences of Rizhao partridge chicken ATP5 A1 W was closely related to goose and mallard. The residing probability of ATP5 A1 W in the mitochondrial, cytoplasmic, nuclear and extracellular was 78.3%, 8.7%, 8.7% and 4.3%, respectively. According to secondary structure prediction, it was composed of α-helix(42.86%), extended strand(19.89%), β-turn(10.49%) and random coil(26.76%). These results will provide data for further investigation on structures and functions of ATP5 A1 W of Rizhao partridge chicken.
