4种菌根真菌对五氯酚耐受性及其生理基础研究

摘  要:
通过纯培养的方法,研究了4种外生菌根真菌——美味牛肝菌(Boletusedulis)、褐环乳牛肝菌(Suillusluteus)、丝膜菌、(Cortinarrusrussus)和厚环粘盖乳牛肝菌(Suillusgrevillei)的生长效应、对五氯酚的耐受性,并采用点试方法对真菌氧化还原酶的产生情况进行了检测。结果表明,4种外生菌根真菌在MM培养基上的生长速率显著高于MMN培养基。4种外生菌根真菌对五氯酚均有一定的耐受性,厚环粘盖乳牛肝菌(Suillusgrevillei)耐受性最高,其生长抑制率显著低于其它真菌。酶测试结果表明,厚环粘盖乳牛肝菌是4种真菌中氧化还原酶活性较强且酶种类多样的菌株,它表现出很高的过氧化物酶、多酚氧化酶和漆酶活性,是一种具有高效降解芳环结构污染物潜力的菌根真菌。4种真菌在生物量和“酸化效应”方面差异较大。厚环粘盖乳牛肝菌(Suillusgrevillei)对五氯酚的高耐受性可能与其产酶特性和“酸化效应”有关。

摘  要:
Ectomycorrhizal fungi has ability to degrade many organi c substances i n nature,so more and more attention h ad focused on how to use the se type fungi to accelerate decomposition of pollutants.In our experiment,the myceliumbiomass or colony diameter of four ectomycorrhizal fungi growing on cult ural media with or without pen tachlorophenol were measured as well as pH value ,oxi-doreductase activities of ectomyco rrhizal fungi Boletus edulis,Suillus luteus,Cortinarrus russus and Suillus grevillei.The enzyme activity was detec ted by spot test.T he results indicated that the four fu ngi grew better on MM medium than MMN medium.Suillus grevillei could grow on the medium co ntained 1 7.6mg ·L-1 pentachlorophenol.At 8.8mg ·L-1 and 17.6mg ·L-1 pen-tachrolophenol levels,the inhibiting ratio of Suillus gevillei growth was significantly lower than other f ungi.Suillus gevillei is the most promisin g fungus that will be used in bioremediation soil polluted by org anic chemicals with phenyl group for its strong oxidoreductase activity produced during its gr owth process.In addition,after culture without pollutant,the pH value of me d ium dropped signifi-cantly,and this effect may play important role in its tole rance and degra dation of Pentachlorophenol.There are significant different ei ther in biomass or in lowering pH of cultural solution among fungi.The h igh to lerant capability to pentachl orophenol may related to their oxidoreductase acti vities an d acidification effect.
