摘 要:
目的:探究不同组分羧甲基茯苓多糖(CMP)对HepG2细胞增殖的影响。方法:在不同提取碱浓度和碱处理时间下,采用二次加碱法制备不同组分CMP。利用硫酸-蒽酮法、滴定法和CCK-8法对CMP的含量、取代度和HepG2细胞增殖检测。结果:获得7个组分CMP,含量为85%~90%,取代度为0.65~0.80。不同浓度CMP培养HepG2细胞24 h下,CMP 750μg/mL抑制细胞增殖效果最好,其中CMP3抑制率最高达50.035%。CMP 750μg/mL培养HepG2细胞24、48、72 h,在48 h下抑制效果最好,其中CMP3抑制效果最高达到76.05%。在72、48、24 h下CMP3培养HepG2细胞的IC50分别为26.34、34.06、763.64μg/mL。结论:CMP能有效抑制HepG2肿瘤细胞的增殖,其中CMP3是7个CMP组分中抑制效果最好的,这与CMP3的含量、浓度呈正相关性,48 h为CMP最适抑制时间。

摘 要:
[Objective] To study the apoptosis of HepG2 cells in different components of carboxymethytlpachymaram(CMP).[Method] CMP of different components was prepared by double alkali addition under different extraction concentration and treatment time.Effects of sulfuric acid-anthrone,titration and CCK-8 on CMP content,degree of substitution and HepG2 cell proliferation were studied.[Result] Totally 7 components of CMP were obtained,whose polysaccharide content was within the range of 85 %~90 % and degree of substitution was 0.65~0.80.CMP 750 μg/mL had the best inhibitory effect on HepG2 cells at 24 h,and the CMP3 inhibition rate was up to50.035%.CMP 750 μg/mL cultured HepG2 cells for 24~72 h had the best inhibitory effect at 48 h,of which CMP3 had the highest inhibitory effect at 76.05 %.The IC_(50) of CMP3 at 72 h,48 h and 24 h was 26.34 μg/mL,34.06 μg/mL and 763.64 μg/mL,respectively.[Conclusion] CMP can induce the apoptosis of HepG2 cells.Under certain treatment time and concentration,CMP can effectively inhibit the proliferation of HepG2 cells,and its inhibition rate is closely related to the dose-effect and time-effect of concentration,as well as the content and degree of substitution of CMP.
