红菜薹游离小孢子培养和植株再生的关键因素研究

摘  要:
对影响红菜薹游离小孢子培养和植株再生的关键因素进行了研究。结果表明,在小孢子胚诱导上,22个基因型之间的小孢子胚诱导率差异显著;对供体材料低温处理后,在0~5 d范围内小孢子胚诱导率差异不大,超过5 d则明显降低;高温诱导在12~60 h范围内差异不大,超过此范围则小孢子胚诱导率明显降低;NLN培养基中活性炭的有无和浓度大小对小孢子胚诱导率的影响极大。在植株再生方面,对胚进行低温处理和提高琼脂浓度能提高胚植株的再生率。

摘  要:
Several factors affecting isolated microspore culture of purple flowering stalk(Brassica campestris L.ssp.chinesis var.purpurea Tsen et Lee) were studied.The results showed that there were significant differences in the frequency of microspore-induced embryoes among different genotypes.For the low temperature treatment,no significant differences had been found during 0~5 days,while the frequencies were obviously lower when the period was longer than five days;there were no significant differences in high temperature when the incubation time was within 12~60 h,but the frequencies of microspore-induced embryoes were much lower over this duration;the presence and concentration of activated charcoal in NLN medium had great influence on the frequency of microspore-induced embryoes.To the embryoes produced from microspore culture,the regeneration frequency was heightened while treated with low temperature or solidified with high concentration of agar in the medium.
