基于重组蛋白TPx的羊肝片吸虫感染Dot-ELISA检测方法的建立

摘  要:
利用大肠杆菌原核表达系统表达的羊肝片吸虫硫氧还蛋白过氧化物酶(thioredoxin peroxidase,TPx)为诊断抗原,建立羊肝片吸虫感染Dot-ELISA检测方法。结果显示:重组蛋白约为30 000;各组分的最适反应条件为:抗原最适包被量为0.28μg/片,血清最佳工作浓度和工作时间分别为1∶400和30min,酶标二抗最佳工作浓度和工作时间分别为1∶800和45 min,血清和酶标二抗最佳反应温度均为37℃,最适封闭液和封闭条件分别为2%BSA-TBS和37℃2h,最佳显色时间30min。该方法特异性较强、灵敏度高、重复性好。运用该方法对浙江海宁和桐乡共200份羊血清样本进行检测,阳性率为1.5%。结果表明:本试验应用重组蛋白TPx通过PVDF(聚偏氟乙烯微孔)膜固相载体建立了羊肝片吸虫Dot-ELISA检测方法。

摘  要:
A Dot-ELISA for detection of sheep Fasciola hepatica was successfully established by the thioredoxin peroxidase(TPx)recombinant protein expressed in Escherichia coli prokaryotic system.The protein was about 30 000.The optimal condition for Dot-ELISA were as follows:the optimal coating concentration was 0.28μg/piece;the optimal serum dilution and incubation time were 1∶400and 30 min respectively,the dilution of HRP-labeled antibody IgG and incubation time were 1∶800and 45 min respectively.The optimal temperature of both serum and HRP-labeled antibody IgG was 37℃.The optimal blocking reagent was 2% BSA-TBS and blocking condition was 2hunder 37℃.The optimal coloration time was 15 min.The results indicated that the established method was highly specific,sensitive and repeatable.The positive rate of 200 serum samples from Haining and Tongxiang in Zhejiang province was 1.5%.
